Job Description
The Proteomics Group at the Broad Institute is a leader in the development and application of targeted, quantitative proteomics technologies (e.g., multiple reaction monitoring, parallel reaction monitoring and data independent approaches). We are looking for a highly motivated PhD-level Research Scientist to join our proteomics team to continue to develop and apply targeted technologies in order to answer pressing questions in biology and medicine. A key feature of this role will be to act as a liaison between discovery proteomics applications and high-throughput, targeted methodologies that will have broad impact for the Broad Institute scientific community.
OVERALL RESPONSIBILITY
Lead and manage multiple complex proteomics research projects of significant scope simultaneously, including independently defining overall strategies and designing complex experiments. Initiates and executes new project concepts.
CHARACTERISTIC DUTIES
• Independently define the overall strategy and experimental design for proteomics experiments including methods for sample handling, protein/peptide fractionation, quantitative mass spectrometry, data analysis and data integration.
• Lead small to moderate sized teams of either direct reports or matrixed reports from within and outside the proteomics group to apply proteomics for biological sample analyses.
• Utilize extensive expertise in MS-based proteomics and biological sample handling for basic biology studies, biomarker discovery and biomarker validation.
• Utilize experience in targeted, quantitative MS methods (including MRM-MS with stable isotope standards) to develop highly multiplexed panels of assays for key proteins and modified peptides to conduct quantitative, time-resolved proteomics experiments for analysis of cell signaling, effects of drug perturbations, candidate biomarker verification, etc.
• Utilize extensive experience in interpretation of quantitative proteomic data analysis including appropriate statistical data analysis expertise
• Work with computational biologists to combine proteomics data with orthogonal data/knowledge sources.
• Serve as primary technical resource and lead for development and optimization of robust, reproducible analytical strategies for targeted MS analysis
• Counsel and train team members on experimental techniques and theories
• Actively participate in the setup of the proteomics laboratory by selecting, procuring and installing, and validating instrumentation;
• Independently design and conduct complex, cutting-edge experiments; compile and analyze experimental data, prepare reports operate and maintain mass spectrometers and HPLC systems.
• Lead and manage multiple, complex proteomics projects of significant scope simultaneously; write necessary reports for internal and external funding/grant agencies.
• Participates in external activities, technical meetings, invited symposia, etc.
• Initiates new project concepts and works with PI to seek funding. Develops technical proposals and makes presentations to funders as required.
SUPERVISION EXERCISED
• Will supervise one or more scientists including post-docs or technicians, provide major input to staffing of project teams, and or mentor / train staff.
• Will lead small teams of either direct reports or matrixed reports from within and outside the proteomics group to apply proteomics for biological sample analyses.
Requirements: • Ph.D. in chemistry or biochemistry and 0+ years of postgraduate experience in mass spectrometry of proteins and peptides and protein biochemistry.
• Relevant experience with HPLC-based fractionation, enrichment and multidimensional LC-MS/MS of complex biological samples, such as biofluids, cell lysates or tissue extracts.
• Hands-on experience with development of quantitative, multiplexed assays by mass spectrometry.
• Strong interest in improving reproducibility, robustness and depth of coverage of current methods for the analysis of signaling pathways and biomarker discovery.
• Strong interest in developing and optimizing relevant new technology for targeted, quantitative, high precision measurement of analytes such as data independent analysis approaches
• Hands-on experience with 1 and/or 2D gel electrophoresis, immunoprecipitation, western blotting, and chemical/enzymatic protein digestion methods.
• Proven team/collaborative skills and leadership capability.
• Excellent verbal and written communication skills.
• Computational and statistics skills necessary for proteomics data evaluation and quantitative data analysis
• Demonstrated leadership and managerial capability of small teams of scientists
• Significant track record of presentation and publication of scientific results at meetings and in peer reviewed journals, commensurate with experience.
• Ability to manage large instrumentation laboratory, overseeing installation and specification testing for highly complex analytical lab instrumentation such as hybrid MS systems.
